A new field of gene expression regulation research is emerging that has previously been overlooked. This new area is concerned with distinguishing the expression of a single gene from the averaged expression of many gene copies within the cell population. This paper reviews research focused on individual genes in inducible gene expression systems. The main experimental strategy is to measure the gene expression level of a single cell containing a single reporter gene molecule. Deep brain stimulation of the subthalamic nucleus is a clinically effective tool for treating medically refractory Parkinson’s disease, but its neural mechanisms remain debated. Previous work has demonstrated that STN DBS results in evoked potentials in the primary motor cortex, suggesting that modulation of cortical physiology may be involved in its therapeutic effects. Due to technical challenges presented by high-amplitude DBS artifacts, these EPs are often measured in response to low-frequency stimulation, which is generally ineffective at PD symptom management. This (...) study aims to characterize STN-to-cortex EPs seen during clinically relevant high-frequency STN DBS for PD. Intraoperatively, we applied STN DBS to 6 PD patients while recording electrocorticography from an electrode strip over the ipsilateral central sulcus. Using recently published techniques, we removed large stimulation artifacts to enable quantification of STN-to-cortex EPs. Two cortical EPs were observed – one synchronized with DBS onset and persisting during ongoing stimulation, and one immediately following DBS offset, here termed the “start” and the “end” EPs respectively. The start EP is, to our knowledge, the first long-latency cortical EP reported during ongoing high-frequency DBS. The start and end EPs differ in magnitude and latency, and the end, but not the start, EP magnitude has a significant relationship to ongoing high gamma power during the EP. These contrasts may suggest mechanistic or circuit differences in EP production during the two time periods. This represents a potential framework for relating DBS clinical efficacy to the effects of a variety of stimulation parameters on EPs. (shrink)

Richard Janko's acclaimed translation of Aristotle's _Poetics_ is accompanied by the most comprehensive commentary available in English that does not presume knowledge of the original Greek. Two other unique features are Janko's translations with notes of both the _Tractatus Coislinianus_, which is argued to be a summary of the lost second book of the Poetics, and fragments of Aristotle’s dialogue On Poets, including recently discovered texts about catharsis, which appear in English for the first time.

In _Isonomia and the Origins of Philosophy_—published originally in Japanese and now available in four languages—Kojin Karatani questions the idealization of ancient Athens as the source of philosophy and democracy by placing the origins instead in Ionia, a set of Greek colonies located in present-day Turkey. Contrasting Athenian democracy with Ionian isonomia—a system based on non-rule and a lack of social divisions whereby equality is realized through the freedom to immigrate—Karatani shows how early Greek thinkers from Heraclitus to Pythagoras were (...) inseparably linked to the isonomia of their Ionian origins, not democracy. He finds in isonomia a model for how an egalitarian society not driven by class antagonism might be put into practice, and resituates Socrates's work and that of his intellectual heirs as the last philosophical attempts to practice isonomia's utopic potentials. Karatani subtly interrogates the democratic commitments of Western philosophy from within and argues that the key to transcending their contradictions lies not in Athenian democracy, with its echoes of imperialism, slavery, and exclusion, but in the openness of isonomia. The United Nations Global Compact (UNGC) was created in 2000 to leverage UN prestige and induce corporations to embrace 10 principles incorporating values of environmental sustainability, protection of human rights, fair treatment of workers, and elimination of bribery and corruption. We review and analyze the GC’s activities and impact in enhancing corporate social responsibility since inception. First, we propose an analytical framework which allows us to assess the qualities of the UNGC and its principles in the context of external and (...) internal elements that influence code effectiveness and implementation. Second, we analyze UNGC performance in encouraging companies to become signatory members and bring about demonstrable change in corporate CSR-sustainability activities. In its 10-year report, UNGC has proclaimed growth in both membership and program activity. However, all credible and publicly available data and documentation conclusively demonstrate that the UNGC has failed to induce its signatory companies to enhance their CSR efforts and integrate the 10 principles in their policies and operations. The result has been a loss of public trust and support of UNGC from important constituencies among civil society organizations, and those individuals and groups adversely impacted by corporate activities and resultant negative externalities. This diminished credibility has also made UNGC largely dependent on the corporate sector for its very survival. We conclude that this dependence has in turn impaired and would continue to hinder UNGC’s ability to fulfill its mission. Such an outcome raises serious questions as to the viability, usefulness, and continued existence of UNGC. (shrink)

Genomic research is an expanding and subversive field, leaking into various others, from environmental protection to food production to healthcare delivery, and in doing so, it is reshaping our relationship with them. The international community has issued various declaratory instruments aimed at the human genome and genomic research. These soft law instruments stress the special nature of genomics and our genetic heritage, and attempt to set limits on our activities with respect to same, as informed by the human rights paradigm. This paper examines the primary thrust and, more importantly, the joint value position of the Universal Declaration on the Human Genome and Human Rights and the Universal Declaration on Bioethics and Human Rights, concluding that, though important legal instruments from the human rights paradigm, these instruments, or rather the values contained therein, must find a more influential hard law voice and a broader policy environment. Utviklingen innenfor informasjons- og kommunikasjonsteknologi har gitt opphav til en type etisk-politisk utfordring som gjerne kalles digital ulikhet. Tradisjonelt har det dreid seg om sosiale ulikheter i tilgangen til IKT på individnivå. Nyere forskning på området er mer opptatt av bruk og dermed av tilbudet av aktiviteter og tjenester gjennom Internett. I denne artikkelen ser vi på hvordan det kan oppstå geografisk digital ulikhet som følge av at norske kommuner tilbyr digitale tjenester – herunder informasjon og ressurser for lokalpolitisk deltakelse (...) – i svært forskjellig omfang. Det betyr at kommunene står overfor etisk-politiske utfordringer knyttet til digital ulikhet som følge av deres strategier for bruk av hjemmesider, og for digital utvikling av kommunen mer generelt. Selv om det er få kommuner som tilbyr særlig mye når det gjelder lokalpolitisk deltakelse, er ulikhetene i tilbudet av informasjon og digitale tjenester betydelige. I tillegg bidrar velutviklede hjemmesider til å gjøre kommunen mer gjennomsiktig og lettere tilgjengelig, noe vi antar er en fordel for innbyggerne. From social to geographical digital dividesThe development of information and communication technologies has raised a kind of ethical–political challenge usually referred to as a digital divide. Traditionally, this has fomented concerns about social inequalities with respect to individual access to ICT. More recent scholarship argues the need to be more focused on use rather than access; accordingly, also on the supply of activities and services through internet. In this article, we study how geographical digital divides may appear as a consequence of the fact that Norwegian local governments supply digital services – including information and resources for local political participation – to quite a different extent. This means that local governments face ethical–political challenges related to digital divides as a consequence of their strategies for the development of home pages and for digital development of local government services more generally. Even though only a few local governments offer much with regard to local political participation, the inequalities with respect to the supply of information and digital services are considerable. In addition, apparently, well–developed home pages make the local government more transparent and more accessible. We believe this to be of advantage to local citizens. The analysis is based on a quantitative content analysis of all Norwegian local governments and a qualitative content analysis of the home pages of 10 local governments. (shrink)
